چکیده

Mast cells (MCs) are tissue-resident immune cells, which play a pivotal role in skin hypersensitivity reactions. MCs activation via Mas-Related G Protein-Coupled Receptor X2 (MRGPRX2) is independent of the classical IgE-mediated pathway, gained much interest recently. Calcium mobilization crucial to mast cell and functional regulation. Ryanodine receptor (RYR) expresses on sarcoplasmic reticulum regulates intracellular calcium liberation. Although MRGPRX2 induces boosted levels, RYR mediated MC has not been fully addressed. We confirmed expression human skin-derived recognized most abundant RYR3 isoform. Human line (LAD2 cells) was utilized study triggered by agonists (c48/80 Substance P). pretreated with inhibitor (dantrolene) suppressed both release extracellular entry stimulated ligands. Degranulation assessed β-hexosaminidase likewise inhibited dantrolene. IL-13, IL-8 CCL-2 induction were at mRNA level when pre-treated inhibitor. The results demonstrate signaling, provide potential approach for MRGPRX2-mediated disorders.

Upregulation of Mas-related G Protein coupled receptor X2 in asthmatic lung mast cells and its activation by the novel neuropeptide hemokinin-1

Hemokinin-1 (HK-1) is a novel neuropeptide produced by human bronchial cells and macrophages and causes contraction of human bronchi ex vivo. It is also generated by antigen/IgE-activated murine mast cells (MCs) and contributes to experimental chronic allergic airway inflammation via the activation of the neurokinin receptor-1 (NK-1R) expressed on murine MCs. We found elevated MC numbers in the...

G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

The Role of Mas-Related G-Protein Receptor X2 (MRGPRX2) on Neuropeptide Induced Reponses in Human Mast Cells

Rationale: Substance P (SP) and hemokinin-1 (HK-1) are neuropeptides (NPs) that promote inflammatory responses by signaling through the neurokinin-1 receptor (NK-1R). Antagonists of NK-1R are highly effective in allergic inflammation and airway hyperresponsiveness in mice but lack efficacy in humans. The reason for this difference is unknown. Human mast cells express both the NK-1R and Mas-rela...

The Mast Cell Receptor Mas-Related Gene X2 (MrgX2) in Health and Disease

Mas-related gene X2 (MrgX2) is a receptor expressed on human mast cells (MCs) but its role in health and disease is unknown. We therefore utilized immunohistochemistry and immunofluorescence techniques to determine the expression of MrgX2 in normal and diseased human tissue. We also tested the feasibility of utilizing humanized mice as a model for in vivo functional studies on MrgX2. We analyze...

Cell-specific extracellular signal-regulated kinase activation by multiple G protein-coupled receptor families in hippocampus.

Several families of G protein-coupled receptors (GPCR) have been shown to activate extracellular signal-regulated kinase (ERK) in transfected cells and non-neuronal systems. However, little is known about GPCR activation of ERK in brain. Because ERK is an important component in the regulation of synaptic plasticity, in this study we examined ERK activation by three families of GPCR that respond...
